研究方向

本研究室著重於幹細胞與癌症領域的探討,並將成果轉譯至再生醫學與癌症治療上。透過與林口長庚紀念醫院臨床資源之整合,我們亦將發展可用於病人之新穎的臨床診斷方法與治療策略。我們利用創新的技術平台以探討人類胚胎幹細胞或腫瘤幹細胞之獨特標記或調節機制。特別的是,我們利用醣化學/醣蛋白體學、幹細胞生物學、誘發性多功能幹細胞等相關技術並結合基因修飾的平台,進而探究各種幹細胞上不同的醣類/醣蛋白之表現,與其下游因子之調控。藉由我們在人類胚胎幹細胞或腫瘤幹細胞上新發現的標記蛋白,以及針對肺部幹細胞之新的分離技術,我們未來的研究將著重於(1)人類胚胎幹細胞或癌症幹細胞的特殊標記分子(glycosphingolipidsN-linked sialoglycoproteinsphosphoproteines)、下游調節因子、與部份精選過的醣轉化酵素在維持自我更新與分化能力之角色,(2)胚胎幹細胞與癌症間的關係,(3)醣質對於幹細胞之功能與分化能力上之潛在貢獻,(4)肺部幹細胞在支氣管分化中之功能與肺氣腫形成的機制,以及第二型上皮-間質轉化在促進小呼吸道纖維化之相關機制。藉由這些新發現的細胞表面標記分子,目前的研究成果將有助於解決幹細胞與癌症幹細胞之分離與增生,也有助於闡明這些幹細胞與癌症幹細胞的新分子在功能上的角色。另一方面,這些嶄新的技術平台發展除了可促進我們更加了解幹細胞與癌症間的相互關係外,也可促進癌症偵測與針對新醣質/醣蛋白療法的發展。
