Research interests:


1. Induction of COX-2, cPLA2, VCAM-1, ICAM-1, and MMP-9 by cytokines, endotoxins, and cigarette extract in airway resident cells.
Several factors have been shown to trigger the pathogenesis of airway inflammation, including IL-1b, TNF-a, endotoxins, and cigarette smoke extract. Recently, oxidative stress due to generation of ROS can also be sensed by the cells and trigger intracellular signaling cascades potentiating chronic inflammation. Cytokines exert as potent stimuli in inflammatory responses through up-regulation of many gene expressions, including cPLA2, COX-2, adhesion molecules and MMP-9. The extent of these gene expressions is correlated with the severity of inflammation. However, the mechanisms underlying the expression of target proteins regulated by these factors are elusive. Therefore, we will investigate the mechanisms underlying the intracellular signaling pathways involved in several target protein expressions induced by these factors in airway resident cells. Increased understanding of signal transduction mechanisms underlying target protein gene expression will create opportunities for the development of anti-inflammation therapeutic strategies.

2. Roles of inducible inflammatory proteins in neurodegenerative diseases
Neuroinflammation is a hallmark of all major CNS neurodegenerative disorders such as Alzheimer's disease (AD) and Parkinson’s disease (PD). Most notably, cytokines (IL-1b and TNF-a), LTA, oxidized LDL, bradykinin and thrombin have been implicated in neurodegeneration. Recently, oxidative stress due to generation of ROS  may be involved in brain injury and inflammation. ROS can also be sensed by the cells and trigger intracellular signaling cascades potentiating chronic inflammation. Interplay between ROS and inflammatory proteins induced by proinflammatory mediators leading to neurodegeneration remains largely unknown. Therefore, we hypothesize that the proinflammatory mediators-initiated neurodegeneration is mediated by the generation of ROS and the induction of inflammatory proteins in brain cells. To confirm these hypotheses, we will investigate the mechanisms underlying BK, IL-1b, LTA, thrombin, ET-1 and ox-LDL-induced expression of MMP-9, cPLA2, and COX-2 rat brain astrocytes.

3. Roles of ATP, Ox-LDL, bradykinin, phospholipids, and thrombin in the pathogenesis of cardiovascular diseases.
Atherosclerosis is a risk factor of morbidity and mobility, which is recognized as an inflammatory disease. Several cytokines and serum factors have been shown to induce the expression of inflammatory target proteins exaggerated the severity of cardiovascular diseases. To elucidate the mechanisms underlying ATP, Ox-LDL-, bradykinin-, phospholipids-, TNF-a-, and thrombin-induced the expression of inflammatory proteins such as COX-2, cPLA2, MMP-9, VCAM-1, and ICAM-1 and PGE2 synthesis in cultured vascular smooth muscle cells and cardiomyocytes, the experiments will be performed to determine the expression of inflammatory proteins and mediators in these cells. An increased understanding of signal transduction pathways involved in expression of these inflammatory proteins may be of potential therapeutic value in the treatment of artherosclerosis and heart failure.

4. The roles of MMPs in tissue regeneration of bone and corneal epithelial cells
Our long-term goal of the research is to elucidate the molecular mechanisms how osteoblasts respond to mechanical loading. Changes in the pattern of mechanical loading will change the internal architecture of a bone organ as an adaptation to the mechanical demand. The process of bone remodeling under mechanical loading may repair fatigue damage and determine bone strength. However, it remains unclear whether mechanical loading directly coupled to specific gene expression and protein synthesis. In response to stimuli, a biaxial stretcher system will be used to introduce strain to cultured MC3T3-E1 osteoblast-like cells, and to dissect out the signaling pathways involved din the expression of MMP-13 in these cells.
Recently, clinical and experimental studies have also shown that ex vivo expanded limbal epithelial cells on AM are capable of restoring the corneal surface with LSCD. However, the underlying molecular mechanisms in this co-culture model are not fully understood. As we know, several extracellular stimuli, including the cell-cell matrix interactions, elicit a broad spectrum of biological responses through activation of MAPKs, transactivation of growth factor receptors, PI3K/Akt, NF-kB, and AP-1 cascades leading to the expression of MMP-9. Therefore, the objective of this study will investigate the regulatory mechanisms of cell migration and proliferation of limbal epithelial cells co-cultured with AM, induced MMP-9 expression through activation of intracellular signaling transduction pathways and transcription factors.
