研究方向:

(1)以蛋白質體學技術平台找尋與驗證惡性腫瘤生物標誌:

惡性腫瘤為人類主要死因之一。大部份惡性腫瘤被診斷時皆為中晚期,是惡性腫瘤死亡率居高不下的主因。除了養成健康生活習慣外,早期診斷與治療是對抗惡性腫瘤的唯一有效方法。雖然有效的惡性腫瘤標誌並不多,抽血檢查其中可能的腫瘤標誌仍是最方便的初步診斷方法。因此找尋有效的惡性腫瘤標誌是對早期診斷惡性腫瘤非常重要的課題。

。以質譜學為基礎之蛋白質體學為近年來新興學門,由於具備高靈敏度、高精確度與高通量之特性,被學界寄予厚望可以用來系統性分析(包括定性與定量)癌症相關樣品中蛋白質,藉以找尋可能的腫瘤生物標誌。敝人實驗室與本校多位同仁合作,共同建立本校蛋白質體學核心實驗室,發展相關定性與定量分析技術平台,分別利用三種生物材料(癌症病患組織體液癌細胞株)找尋癌症生物標誌及分析癌症之異常分子調節機轉。

(2)蛋白激脢PAKGSK-3之生理功能及調控機制:

可逆的蛋白質的磷酸化-脫磷酸化作用是一種調節細胞諸多生理功能的重要機制。蛋白激脢酵素群(protein kinases)負責蛋白質的磷酸化,而蛋白質脫磷酸化酵素(protein phosphatases)則負責脫磷酸化作用。生物體透過這兩類酵素活性在體內所達成的特定平衡狀態,來調節體內各式各樣的關鍵酵素和蛋白的磷酸化程度,迅速而有效的改變它們的構形與活性,進而對外在刺激作出適當反應。因此,這兩類酵素群在生物體內訊息傳遞作用機制中扮演相當重要角色。本實驗室目前針對兩種蛋白激脢—p21-activated kinase 2 (PAK2)glycogen synthase kinase-3 (GSK-3)分別在細胞凋亡(apoptosis)及癌化過程中可能扮演之生理功能角色及調控機制作深入探討。研究重點在於(A)利用蛋白質體學方法找尋此兩種蛋白激脢的結合分子與下游受質;(B)找尋與發展此兩種蛋白激脢的小分子抑制劑

(3)光照療法對癌細胞的影響-細胞功能變化與分子調節機制:

光照療法(photodynamic therapy, PDT )屬於光化療法的一種,這種療法已通過許多國家包括美國之衛生藥物檢驗相關單位核可,用來治療部分癌症。新陳代謝速率較高的組織(特別是腫瘤組織)對光敏劑(photosensitizer)的吸收能力較好;光照療法即是利用光敏劑此一特性,利用特定波長光線對已經吸收光敏劑之癌細胞組織照射,由於光敏劑吸收特殊波長光能後,會將能量轉換給組織內氧分子,促使高能含氧活化物(reactive oxygen species, ROS)生成,隨後引發細胞中一連串的訊號傳遞,進而導致癌細胞死亡,而達到治療效果。本實驗室曾分析人類皮膚癌A431細胞經過Rose Bengal (一種光敏劑)-PDT處理後,所引發的細胞凋亡訊息傳遞機制。目前我們與長庚醫院整形外科合作,進一步探討臨床光照療法對癌細胞影響之分子作用機轉;我們使用臨床用藥Photofrin®及氦氖雷射對癌細胞進行PDT處理,發現Photofrin®可以分佈在癌細胞內部不同的位置,而隨著分佈的位置不同,PDT所引發的死亡型態與機制亦不相同。其詳細分子機制正在探討中。

研究室特色

實驗室累積多年蛋白質酵素分離純化與鑑定分析之經驗,以瞭解數種蛋白激脢及蛋白質脫磷酸化酵素之分子活性調控機制為出發點,進一步利用各種生化、分子生物、免疫、組織細胞培養及細胞生物學技術,以訊息傳導為核心,試圖由多重角度來探討幾個我們感興趣的生命科學課題。研究內容生動活潑,蘊藏無限可能性應是本研究室最大特色。歡迎對基礎醫學研究充滿熱誠之同學加入我們的研究團隊。
