This lab aims to investigate relationships and interactions between flaviviruses and their host cells. Flaviviruses has been known to cause a variety of human diseases including dengue fever and Japanese encephalitis; both are prevalent in Taiwan. The genome is composed a positive polarity single-stranded RNA which readily alters during transmission between host cell, frequently resulting in changes of the phenotype of the virus such as infectivity and pathogenicity. According to our previous studies, genetic variation of the viral genome may be caused by mutation(s); besides, RNA recombination actually also plays an important role in this feature. Most flaviviruses are naturally transmitted between mammalian hosts by blooding sucking arthropods such as mosquitoes. As a result, they are usually capable of infecting both mammalian and arthropod cells. Generally, infected mammalian cells end up with apoptosis while most mosquito cells can survive the infection. It indicates that interactions and cellular responses to the viral infection must be significantly distinct between the cell types. In the meantime, a novel tetraspanin was recently identified and demonstrated to involve in cell-to-cell transmission of the dengue virus between cells. All these interesting topics are currently worked in this lab via molecular and imaging techniques.
