  1. Oral cavity squamous cell carcinomas (OSCC), the fourth most prevalent among males in Taiwan, is an aggressive tumor and is the most common malignancy in the oral cavity, representing up to 90% of all malignant neoplasms of the oral cavity. The highly recurrence rate and metastasis is still major problem for OSCC treatment. We previously identified and published the specific genetic signatures in OSCC patients from Taiwan using whole exome sequencing. Furthermore, we performed the RNA-seq analysis, quantitative reverse transcription polymerase chain reaction (RT-qPCR) assays, and immunohistochemistry (IHC) staining of tumor tissues and adjacent non-tumor tissues from OSCC patients for characterization and validation of the potential dysregulated genes and proteins in oral cancer. Thus, development of target therapy with precision medicine for OSCC are urgent need. Also, no useful genetic biomarkers are reliable for OSCC patient selection. Patient-derived oral cancer xenografts (PDX) maintain the molecular, genetic and histological features of origin patients’ tumor through serial passaging in mice. The PDX models can provide a more-relevant system to test clinically directed questions. Our lab established and characterized the histopathological features and genomics signatures in paired primary tumor and xenografts. We intend to clarify the useful biomarkers for predicating drug response and mechanisms in oral cancer.
  2. Parapneumonic effusion (PPE), which occurs approximately in 40% of pneumonia patients, is an accumulation of exudative pleural fluid associated with the pulmonary infection. The morbidity and mortality of pneumonia increase when the patient presents with the PPE, because this stage correlates with more-advanced pneumonia. Based on fluid characteristics, PPEs are classified as uncomplicated parapneumonic effusions (UPPE), complicated parapneumonic effusions (CPPE), or empyema. The pneumonia patients with empyema have a 50% mortality rate. A number of potential immune-related proteins have been evaluated in our lab. We aim to identify potential biosignatures to aid in distinguishing different stages and predicating outcomes of PPE.
  3. The high-throughput sequencing technology is becoming a dominant platform for the study of microbial population genomics. With this culture-free and cost-effective technique, researchers can combine multiple samples in a single sequencing run, studying taxonomic composition of samples from environments, complex microbiomes or clinical specimen. The variable regions of bacteria 16S rRNA are frequently used in phylogenetic classifications to identify genera or species. High-throughput 16S rRNA sequencing technology provides more comprehensive bacteria profiling for revealing hundreds of bacteria in clinical samples, which can facilitate the discovery of bacteria biomarkers. Our lab established the microbiome sequencing platform and bioinformatics tools for identifying the microbiome community in various clinical samples such as saliva, sputum, and stool. We aim to identify the correlation of microbiome and disease progression.
