研究室特色:

分子調控暨生物資訊研究室之主要研究目標為以單細胞生物陰道鞭毛蟲作實驗材料,探討細胞壽命延長及化的調控機轉。研究特色是利用生物資訊整合各種經由高通量分析平臺所產生之數據對基因體、轉譯體,蛋白質體作為基礎,並從此等資料中進行探磡,發掘可能調命單細胞原生動物壽命延長及化的機轉。

研究方向:陰道鞭毛蟲微小蛋白調控缺鐵反應之研究

微小蛋白質是由少於一百個胺基酸所組成的蛋白質,最近的研究顯示這群蛋白質參與許多細胞內反應,如腫瘤生成、壓力反應及訊息傳遞等。然而,因為傳統的基因註解方式將一百個胺基酸的長度作為一個閥值,將轉譯出短於一百個胺基酸的開放閱讀框排除,導致微小蛋白一直是被忽略的一群。

基於近年發展成熟的次世代定序技術,其應用廣泛,不僅讓微小蛋白質得已被鑑定出來,更藉此能獲得更全面的基因表現資訊。然而截至目前為止,微小蛋白質尚未在任何原蟲中被發現。本研究室將利用陰道鞭毛蟲作為一個模式生物,鑑定並探討微小蛋白質在缺鐵壓力下所扮演的角色。陰道鞭毛蟲是造成人類非病毒性、盛行率最高的性接觸傳染病陰道鞭毛蟲症的病原體。鐵離子對於調控陰道鞭毛蟲細胞內能量代謝、氧化還原平衡及氫化酶體質量等相當重要,因此陰道鞭毛蟲如何在缺鐵情況下生存是相當重要的課題。經由全基體開放閱讀框、第二代/第三代高通量定序及蛋白質體學整合性生物資訊分析,已初步確認在陰道鞭毛蟲的轉錄體及蛋白體中可發現新的微小蛋白。這初步研究結果開啓了一個全新的研域。因此未來計畫的目標為研究陰道鞭毛蟲的微小蛋白質在缺鐵情況的功能性角色,並且建立一套針對微小蛋白質的實驗及分析平台。
