在基因全面解碼的後基因體時代,蛋白質體學分析技術也迎來飛躍的成長。本研究室著重於以先進蛋白質體學定量技術分析生物樣品間的蛋白質差異表現,探討腫瘤的早期診斷、治療標靶、預後評估的相關蛋白質分子。

技術開發: 1) 結合免疫學及蛋白質體學,開發以抗胜肽單株抗體(anti-peptide antibodies)富集樣品中的待測目標胜肽,並利用質譜學多重反應監測技術(Multiple Reaction Monitoring, MRM),進行多重標靶精準定量,稱之為「免疫-多重反應監測技術」(immuno-MRM),應用於臨床唾液及血液樣本中準確定量多種低含量的蛋白質標的分子。

2) 建立目標特異性核酸適體(DNA/RNA aptamer, single stranded DNA/RNA specifically bind to target)之體外選殖技術(systematic evolution of ligands by exponential enrichment, SELEX)平台並發展以核酸適體為基礎之癌症檢測平台。

口腔癌研究: 利用MRMimmuno-MRM技術於唾液檢體中驗證能夠區辨口腔癌病人的蛋白質生物標記組(Protein biomarker panel),建立唾液蛋白質分子的個體差異及日常生理變化之定量化參考資料,進一步驗證生物標記分子在血液及唾液檢體的檢測口腔癌效力,提供實驗數據證實唾液作為常規檢體用於檢測口腔癌的潛力。

腫瘤免疫研究:探討免疫系統在腫瘤發生與惡化的各階段中所扮演的角色,包括: 1) 慢性發炎反應的致癌機轉;2) 腫瘤細胞與免疫細胞間相互作用機制;3) 腫瘤免疫學研究於癌症治療、監測以及抗腫瘤疫苗發展之應用。
