Research interests:
In the post-genomic era, many advanced technologies have been developed for the proteomic approaches after the whole genome sequencing completed. Our researches focus on the differentially expressed proteins discovered in biological samples by using advanced quantitative proteomic approaches. We are trying to explore the particular proteins involved in the early detection, therapeutic target, and prognosis in tumors.
Assay development: 1) We have developed a multiplexed and quantitative platform that combines immuno-affinity enrichment and mass spectrometry, known as “SISCAPA-MRM” (stable isotope standards with capture by anti-peptide antibodies coupled with multiple reaction monitoring) or “immuno-MRM”, and further applied these assays to quantify dozens of low abundant protein targets in saliva and serum/plasma samples. 2) we also established the platform of systematic evolution of ligands by exponential enrichment (SELEX) to generate the target-specific DNA/RNA aptamer, single stranded DNA/RNA specifically bind to target, and will further develop aptamer-based assays for cancer detection.
Oral cancer study: Using MRM and immuno-MRM technologies, we verified a panel of protein biomarkers that capable to discriminate OSCC patient from health. Additionally, we illustrated the personal variations and diurnal fluctuations of salivary proteins and demonstrated the clinical utility of potential candidates in saliva rather than serum/plasma sample. Our study offers a practical foundation for using salivary protein biomarker panel to enable early detection of OSCC.
Tumor immunologic study: To explore the role of immune system in tumorigenesis and progression, the studies include several domains: 1) the carcinogenic mechanism involved in chronic inflammation; 2) the interactions between tumor cells and immune cells: 3) applications of tumor immunology in treatment, monitoring, and anti-cancer vaccine.
