本實驗室以過敏相關疾病為研究主軸,研究疾病的分子機制及開發新療法。過敏與氣喘疾在已開發國家盛行率愈來愈高,病嚴重影響人民的健康與消耗許多醫療資源,這類疾病是由多個遺傳與環境因子引起體內免疫系統不平衡反應所致,我們的研究重心主要在於深入探討可以調控過敏病的分子與細胞間反應,期許應用於研發新的防治方式。

氣喘病之基因治療:基於氣喘常為第二型T 輔助細胞過度反應所致,嚐試利用病毒載體攜帶抗發炎分子或第一型T 輔助細胞相關之細胞激素基因,經由氣道施予疾病模式小鼠,用以平衡過度活化的第二型T 輔助細胞免疫反應;或是攜帶可干擾第二型T 輔助細胞反應的基因片段,評估其抑制氣喘疾病各項指標的可行性。

藥物或是抗體引發T 調節細胞的發展:近年,如何調節過度活化的免疫反應成為免疫學的熱門課題,我們亦著手研究與表觀遺傳學的基因表達調控相關藥物、或是可改變T 輔助細胞族群活化功能的特殊抗體等,對人類周邊血球中的被激活T 輔助細胞或是氣喘疾病模式小鼠的免疫調節功能。在逐步研究這些藥物如何調控氣喘疾病發生的分子機制過程中,可以進一步發展運用這些藥物或抗體來治療氣喘疾病。

探討可治療氣喘中藥之科學證據:以臨床使用療效佳之方劑如「定喘湯」,利用氣喘動物模式來研究中藥使用之劑量、給藥時機、療效機制及安全性,做為分離出有效成分之基礎;也和中、西醫臨床醫師合作,完成療效評估。在中醫藥科學化方面,則將注重中藥作用於細胞訊號傳遞系統之機制,並搭配以中草藥材中的純化藥物,來探討並分析各藥物對周邊淋巴球細胞活性的調控或是對氣喘疾病模式小鼠的抑制發炎功能。

臍帶血淋巴細胞活性研究:比起骨髓,移植臍帶血中造血系統,有材料取得的方便性與排斥性低的優點。所以本實驗室的另外一個研究主題,是和長庚兒童醫院醫師合作,探討以不同細胞激素組合對臍帶血中 T 輔助細胞及自然殺手細胞之複製、死亡及活性的調控,或是評估不同免疫抑制劑對細胞功能的影響,可以對臍帶血移植時的免疫反應,有更多的了解。
